摘要

本文以国内广泛应用的抗甘蔗花叶病毒玉米自交系黄早四为材料,利用cDNA-AFLP差异显示方法,分析其接种病毒后的基因表达差异模式。用56对引物扩增出203个基因差异显示片段,其中187个是诱导性表达,16个是抑制性表达。经反向Northern确认随机挑选的24个基因差异片段中有11个是病毒侵染后诱导表达的,其中有10个片段在GenBank数据库中可以找到显著同源序列,这些基因差异显示片段分别与玉米磷酸丙糖转移蛋白(TPT)、转录调节因子、G蛋白、锌指结构域、果糖1,6-二磷酸酶、芳烃受体等基因有较高同源性。

Abstract

cDNA-AFLP was applied to analyze the differential expression induced by SCMV-BJ in a resistant maize inbred line Huangzaosi which was widely grown in China. There were 203 fragments from the amplification with 56 primer-pairs that showed differential expression profiles between SCMV-BJ infected and control healthy maize plants, among which 187 fragments were induced and 16 inhibited. Among the 24 fragments randomly selected, 11 fragments were confirmed by reverse Northern as inducible by SCMV-BJ. After sequence analysis, 10 fragments were found to be highly homologous to GenBank sequences, including maize triose phosphate translocator (TPT), transcription factor, GTPase regulator, zinc finger domain, aromatic hydrocarbon receptor, fructose-1,6-bisphosphatase.
