Cloning of an ADP/ATP carrier protein-coding genein Blumeria graminis f. sp. tritici and analysis on its expression pattern during infection process

Abstract:Wheat powdery mildew is an important disease in production. It is important to investigate molecular mechanisms of pathogenicity\|related genes in the pathogen Blumeria graminis f. sp. tritici (Bgt) for discovering potentialanti\|fungal targets. Based on our previous transcriptomic data, one ADP/ATP carrier protein\|coding gene(code Unigene0005672) was found to be up\|regulated during the interaction between Bgt and wheat. A cDNA fragment containing a 945\|bp ORF was obtained with RACE. This ORF was tentatively named as BgtAACPx(GenBank accession: MF197857) and encodes a deduced 314\|amino acid protein, which is predicted to contain six transmembrane domains and localize on the mitochondria and have 97% similarity with a known ADP/ATP carrier protein of Bgt. The QPCR result showed that BgtAACPx was highly expressed in haustorium, secondary hyphae, and spore production without forming sorus, suggesting that this gene might be involved in the fungal pathogenicity.
